Yes and no. Madonna was (and is) super smart and funny but I've read stories where she could be really rude and hurtful to people but at the same time, imagine being the biggest popstar in the world and everyone wanting something from you. You would have to have your guard up all the time since you never know when someone is sincere or wants to exploit you.

The post in the Reddit thread came from The Sun… which I’ve heard isn’t always the most reliable source. If it is true and it happened a long time ago, Madonna might have looked at a random person like “why are you breathing my air?” And that wouldn’t be out of character. These days, I can’t see it happening though. 
 

Edit: This was in the 80’s. Ru was working as a GoGo dancer and obviously wasn’t nearly as well known. Not saying what she did was right but to be fair, if some random employee wandered into my room I’d wonder what was going on too.

OT: RuPaul is obviously just trying to drum up some publicity to sell his book and using Madonna's name (especially negatively) is a tried and tested way to achieve that (see Boy George).
